Multi-color two-photon microscopy is a powerful tool for the detection of cellular events in vivo. We propose a dynamic segmentation method, which works well with an existing image contrast between cells and background that are refractive to a reference wavelength. The proposed method segments the cells in the images at different states, then uses the variation between the segmented images to calculate the cell positions in the overlapping regions. The calculated position can be used to define the end points of the segmented cells, and obtain the cell boundary. Finally, the cell boundary is processed to obtain the segmentation results. This method is dynamic, adaptable, and more stable than traditional segmentation methods. It is not only applicable to multi-color two-photon microscopy, but also to other optical microscopy that forms brightfield or darkfield images. In this paper, we first introduce the principle and step-by-step analysis of the algorithm, then illustrate and evaluate the performance of the proposed method.
